PERTUMBUHAN KULTUR MERISTEM MAHKOTA NANAS (Ananas comosus (L.) Merr) PADA MEDIA MURASHIGE SKOOG (MS) DENGAN PENAMBAHAN EKSTRAK JAGUNG (Zea Mays (L.) DAN NAPHTHALENE ACETIC ACID (NAA)

Abstract


Pineapple (Ananas comosus (L.) Merr.) is a kind of horticultural fruit that has able to be cultivated and has good economic value in West Kalimantan. Propagation of pineapple plants can be effort in vitro through pineapple crown meristem culture by growing explants on Murashige Skoog media with the addition of corn seed extract and Naphthalene Acetic Acid (NAA). The research aims to determine the effect of corn seed extract and NAA on the growth of pineapple crown meristem culture. The study was going on September 2019 - April 2020 at the Tissue Culture Laboratory of Biology, Faculty of Mathematics and Natural Sciences, Tanjungpura University. The research used completely randomized design with two factors. The first factor was corn seed extract with a concentration of 0% (J1), 10% (J2), and 15% (J3); the second factor was NAA concentrations of 0M (N1), 5x10-8 M (N2), and 10-7 M (N3). Each treatment was repeated 3 times in order to obtain 27 experimental units. The results showed that the combination of NAA and corn seed extract had a significant effect on bud emergence time (days), number of buds, number of leaves (strands) and plantlet height (cm). The treatment of 10-7 M NAA + 15% corn seed extract (N1J2) produced the best growth with an emergence time of 8.6 days and 7 shoots.
